Usually not on their own, and that is deliberate. A spelling mistake in a passenger name is not really a change in your portal, it is a change at the airline or the hotel, so most portals let the agent raise the request but keep the actual amendment in your hands or your operations team's hands.
The reason is that the supplier decides what is possible, not the software. Some airlines allow a small correction of a few letters free of charge, some treat any name change as a cancellation and a fresh booking, and hotels are often more relaxed than airlines about the same mistake. Because the answer changes from one supplier to the next, and sometimes from one fare type to the next, a portal that let every agent press a button and promise a correction would be promising something it cannot deliver.
What you get instead is a proper trail. The agent raises an amendment request against the booking, it lands with your team, and whatever happens next, the correction, the fee or the refusal, sits on that booking where both sides can see it later. That matters when the agent tells the traveller one thing and your team remembers another. Whether your agents can raise these requests themselves or have to email you depends on how your portal has been set up and on the supplier connection behind that product.
